Mechanism of action of immunosuppressive agents

Ciclosporin and tacrolimus are both calcineurin inhibitors - the former binding to calcineurin as a complex with cyclophillin A, and the latter as a complex with FKBP1.

Sirolimus also binds to FKBP12, but this complex inhibits cell proliferation by binding to the mammalian target of rapamycin (mTOR).



Mycophenolate mofetil, not azathioprine, acts through inosine monophosphate dehydrogenase inhibition.
